Test PCR of Heme Oxygenase Biobrick, entire template used, with 200pg primer and annealing temp reduced to 53.9. Other program left constant.
Result
No bands seen, except for positive control.
- check if DNA is present, give for nanodrop analysis.
II)
Transformation.
Procedure repeated with Sigma kit, yesterdays transformation successful.
III)
Revived cultures of DB3.1 and NEB10b cells.
Inoculated DH5a, DB3.1, NEB10b for transformation tomorrow.
IV)
Brainstorming continued, construct refined a bit.
