I) Transformation with PCR product, dilute DNA 1:10 and use 4ul.

NO Transformants  :-(

Why?

  • Dilution?
  • Linear DNA, try ligation before transformation.
  • Kan conc too high?


II) Ligation test. RE digest of pBR322 and pGFP, gel and ligation.
