Rehydrate R1051 (plate 1, 8E)(amp), P0151 (amp), K145303 (amp) Rehydration Protocol

Retransform R0051 (kan), R0010 (kan) Transformation Protocol


Incubation started at 11:40, Amp parts done at 1:05, Kan parts done at 1:40.




Layne 10am

-Picked colonies from last week's plates: ccdB resistant strain, P1010 Amp backbone plasmid and two I0500 plates (B:original transformation and A:remainder spun down and replated)

-Inoculated each into about 50 ml LB and put in shaker at 37 degrees.


The part I0500 is on an inducible copy number Kan resistant plasmid. The origin of replication is maximally induced in the presence of 100umol/L IPTG


-Added in 20ul of IPTG to each of two cultures of I0500, left to grow overnight.
