Lab work
ON culture
- tri strain (for Caroline) 6ml LB
New transformation
- pSB2K3 in order to have new clones because I can't get the right digestion profil
- NB: 1h20 of incubation at 37°C under agitation (Miam time --!)
PCR
- For A11 (ex A4) ??name??: Matrix = Genomic DNA of K12, Oligo = O11 and O12
- For A13 (ex A5) ??name??: Matrix = Genomic DNA of K12, Oligo = O1 and O13
- 1:98°C 30s
- 2:98°C 10s
- 3:70°C 30s
- 4:72°C 8s
- Goto 3 29x
- 5:72°C 10min
- 6: 4°C ~
NB:1h at 98°C because I haven't push the "enter" buton after it warm up to 98°C...and all evaporate.
