Ex 9: Miniprep of Spinach cDNA did work – digested and undigested gels Gel:

Ex 13: PCR of Choline, ProU, YhfR, nudF

Results: Ran Gel of PCR Product - no DNA

Reasons:

1. I only loaded 1 micro liter out of 50 micro liter, so the concentration may be low to detect;

2. We ran the 0.7% gel too long, since three of the gene of interest are smaller than 800 bps, they may ran into the buffer. We stopped when the dye front was at the edge of the gel.

3. Two of the tubes (yhfR and nudF) got "smashed," but they were not broken

4. Most Likely PCR Cycle did not work


Ex 11: Started Salt Tolerance tests using plate reader

-3 samples of each of the 11 concentrations were tested and placed randomly about the center of the plate (iffy readings on edges)
