Literaturrecherche zu FokI.Entstandene Ideen:
- drei Oligo's: zwei für die Bindung der Monomere und eines zur Erzeugung der dsDNA an die FokI binden kann.
- zwei Oligo's: eines zur Bindung des einen Monomers, das zweite zur Bindung des zweiten Monomers und zur Ausbildung der dsDNA (ein Monomer müsste hierfür inaktiviert werden).
- ein Oligo: beide Bindestellen der Monomere auf einem Oligo. Auch hier müsste ein Monomer inaktiviert sein. Es wurde die 196 C-terminalen Aminosäuren von FokI verwendet. Primersequenzen sind vorhanden.
