ON starter cultures continued for creating biomass for As uptake experiments combined with promotor activation and methalotein binding. Use E. coli + control vector, E. coli + pArsR-RFP, E. coli + pLac-fMT Take OD600 samples every 1-1.5 hrs of E. coli + pLac-fMT. Induce E. coli + pLac-fMT at OD600 ~0.6 with 0.5mM IPTG.
