- Transformation of competent E.coli strain TopF' with ligation.
- Plating ligation on LB+X-Gal+IPTG+Amp plates and leaving for incubation in 37C degree.

- Prepare the bacterial cultures for isolation of following construct:
- mitochondrial signal peptide from Cox CDS on pKSII plasmid

- isolation of previously described construct
Methods:
- Plasmids were isolated using the A&A plasmid mini kit. Detailed procedure of the isolation is described here
Inverting PLacI regulated parts of the switch and assembling the switch on a single plasmid
