Wet Lab

Result of the overnight incubation: Only one or two colony on 4 plates.... The positive control of the transformation grew very well.

Test of other ligase (Quick ligase and other T4 ligase) on LacI-RBS plasmid and GFP into two different plasmid.

Result after an overnight incubation: Nothing grew

Conclusion: We have problem with our ligation. So we need to test it without Phosphatase, with precise concentration and try a gel extraction.
