8.30am

Aliquoted 1.5mL into epis, and left in shaking incubator (it's back!) so they can get a bit of shaking action. I'll be back at 10.30 to do the rest of the miniprep. If anyone is free to help out it would ensure I finish on time to get to my class at 11.30.

12.01pm

Miniprep is all done.

NOTE: Make sure to time the centrifuges always! The one without paper on the top goes way longer than 10 minutes when you set it!

I'll be back after 2.30 to do the gels. I have to be gone at 7.30 tonight, but if these gels work and we're efficient, we should have time to do new assemblies, even if we don't have time to transform them.

3.30pm

Run gel on K235019, 20, 21, plus doubles of J23070, E0240, C0012, K115017, R0051.

Gel started 4.30pm. Gel put in EtBr soak at 5.50pm.

We had no bands show up in our gel.

We will subculture on sunday to have new cultures to miniprep on monday. We will also subculture K235014 to test at different temperatures for GFP on monday.

Troubleshooting:

- Let's try bigger tubes next time.

- It wasn't shaking last time.

- We had satellite colonies on our plates.
