-Received Sigma shipment of tubes, reagents, and restriction enzymes

-Made TAE buffer for electrophoresis gels

-Ran PCR-amplified biobrick parts (from yesterday) on 0.8% agarose gel, to check size

-Visualized gel

-Gel layout:

Lane 1 (from left): 1 kb ladder

2: Sample A: BBa_I15010, red light sensor, 2238 bp

3: Sample B: BBa_R0082, OmpC promoter, 108 bp

4: Sample C: BBa_B0015, terminator, 129 bp

5: Sample D: BBa_J04450, RFP

6: Sample E: BBa_B0034, RBS, 12 bp

7: Sample F: BBa_E0240, GFP, 976 bp

8: Sample G: BBa_J13002, TetR, 74 bp

9: 10 kb ladder


Recipe for loading:

1 μL loading dye + 8 μL sample per lane

105 volts
