We going to digest L2-3 (BBa_K266000) as insert XBAI and PSTI 17 hours digest in 37°C camera.

06-October-2009

The digest L2-3(BBa_K266000) doesn´t work, we have to repeat it. Left 17 hours digest.

07-October-2009

We run out the digest and it is successful. Next step is attach to (BBa_K266006) as backbone. Left 17 hours ligation overnight.

08-October-2009

Transformation by head shock, plate and left overnight in 37ºC.

09-October-2009

The transformation was successful we get five colonies.

12-October-2009

Left two colonies in LB medium inoculums overnight to prepare midi tomorrow.

13-October-2009

We have culture and prepare midi prep.

15-October-2009

Biobricks sent to the registry and final ligation in progress.
