  • PCR'd mCh.1 and mCh.2 using gradient PCR - FAILED
  • Made overnights of pKD46 in MG1655 for making electrocompetent cells
  • Checked miniprepped mCherry (cut with XbaI) for validity
  • Colony PCR'd orn-pBA to test if ligation worked
