The overnight culture of R0051 seems to be very nicely grown; when the most recent batch of goodies gets transformed and needs miniprepping, this one can get done too. I know that I'm interested to try running a repeat gel on the first minipreps of R0051.


Today we got two sleeves of petri dish plates from Stores. Now we need to make some more LB+antibiotic plates - where did we get the stuff before? Central Services, one of the Micro labs?


At any rate, Dr. Upton had some premixed LB+agar that we can use, so we may yet get plates done today!


A bottle of about 750mL premixed LB+agar, heat initally at medium (50%) power in a microwave for quite a while, check periodically. The settled-out agar will take a while to reheat, but on the other hand you don't want to boil the top portion.


250mL for ampicillin, 250mL for chloramphenicol, about 250mL for kanamycin

--> 1.25mL ampicillin stock, 250µL chloramphenicol stock, about 625µL for kanamycin


Also, apparently the promised money hasn't yet been entered into our Stores account so we're technically down $300, although the secretary wasn't worried because she does know we're supposed to get money.
