I can come in tomorrow afternoon at 4.30 to start broth cultures for these.  (Reminder to autoclave epis!  They will be needed for the miniprep the next day!)  And on Thursday I can miniprep these at 8.30am.  If no one else can be here, I can run a gel at 2.30 on Thurs but it would be great if anyone else can do it that day.

Alright, the plan for today is to do the assemblies K19, 20 and 21. Rehydrate the new ribokey J23070, and retransform R0051, E0240, C0012 and K115017.


7pm

- Assembled K235019, 20, 21.

- Rehydrated J23070 (new ribokey), J04630 (GFP + stop: to use as a control)

- Transformed all of the above

- Plated all of the above plus new streak of R0051 stab (I'm not sure what kind of growth to expect from that.)
