Linearizing BioByte Constructs by I-SceI Digestion

What you will need:

  • I-SceI
  • Circular Biobyte Construct

Digestion:

For a 20 μL reaction:
  • 5 μL DNA sample
  • 2 μL NEB 10x I-SceI Buffer
  • 2 μL 10x BSA
  • 10.5 μL ddH2O
  • 0.5 μL I-SceI (5U/μL)

Incubate at 37ºC for 1 hour
