Project 3 is concerned with developing a system which is naturally competent. This is effected by introducing special com genes taken from naturally competent strains of Acinetobacter and Streptococcus into a suitable vector.
Induction of competence will thus enable the bacteria to take up naked DNA from its surroundings which will greatly facilitate transformation experiments.
We also plan to introduce a lytic cassette which, when introduced into a subpopulation of cells will induce their lysis and also subsequently spread to the rest of the population thereby giving rise to a chain reaction of lysis events.
This has a lot of important applications in 1) Preparation of DNA,plasmid 2) Extraction of protein 3) Clinical applications like in DNA vaccines etc.
