CD4 fused with C-terminal half of split-ubiquitin system and T7 RNA polymerase with NLS (nuclear localisation sequence). When assembled with N-terminal half of ubiquitin, the T7 RNA polymerase is cleaved off and translocated into nucleus, where it transcribes genes under T7 promoter.
Due to T7 promoter the expression of the gene is constitutive in mammalian cells.
