1.before Min prep
Checked the cell cultures transformed and plated out yesterday (8/10) for colony formation and made an approximate count of the number of colonies.

        ③                     10     (Amp)
        ④                     50~   (Amp) 
        ⑤                      1     (Kan)
        ⑥                    ~10    (Amp)

4 colony (each ③,④,⑥) inoculate to iquid medium(5mL + Amp 5uL)

2. 0.1M MgCl₂, 0.1M CaCl₂ buffer
