1x TAE Buffer

  • 242g Tris base
  • 57.1mL glacial acetic acid
  • 100mL 0.5M EDTA (pH 8.0)
  • 1L dH2O

First, prepare a concentrated 50x stock solution of TAE buffer. To do this, dissolve Tris base in 750mL of deionized water. Add the acetic acid and EDTA, and adjust the volume to 1L by adding water. The final pH of the 50x TAE buffer should be about 8.5. To make the 1x TAE working buffer, add 49 parts of deionized water to 1 part of 50x TAE buffer.
