Restriction Reaction

- 1X Reaction Buffer

- Total DNA - 0.1-4 μg

- Endonuclease - 1 unit per μg of DNA

- Autoclaved distilled water to 20 uL

Temperature - 37°C - or the suggested by the manufacturer

Time - 3 h or overnight

Protocol according to Current Protocols in Molecular Biology
