I) Gel elution. (Kit used, Bangalore Genei)

A280 of eluted DNA

~500 ng DNA !

Too good to be true, running a gel to confirm.

II) pBR322 isolated successfully.

III) Ran a PCR with Vf and Vr.

Gel positive.

DNA is present, QED.

Try P1 and P2, lets see if a gradient PCR will solve our problem...
