- Grid plates containing mutant 5 promoter constructs were screened using a fluorescent camera to identify which colonies showed the most intense fluorescence.

- 2 mL LB antibiotic cultures were innoculated from the greenest colonies and allowed to grow overnight at 37 °C with shaking at 220 rpm.
