Results

Afton and Maria

  • Electrotransformation was successful
    • Control (+) NEB 10 beta on LB has lawn growth
    • Control (+) NEB 10 beta on LB electroshocked has lawn growth
    • Control (-) NEB 10 beta on AMP has no growth
    • J23106 (promoter) on AMP has hundreds of little colonies
    • J06702 (RET) on AMP has about one hundred colonies

  • Pick cells for overnight culture

Kevin and Adam

  • Proceed with analytical work and ordering DNA.

Craig and Clay

  • The three parts for limonene synthesis (BBa_I742111, BBa_K118024, and BBa_K118025) will be electrotransformed today. They will also be plated and grown overnight on ampicillin-selective media.
  • Tomorrow colonies will be selected and grown in ampicillin-selective liquid media during the day. Later in the evening a miniprep will be performed on the grown parts.

Wetlab

Afton and Maria

  • Put 5 colonies of J23106 in 5mL of LB (two test tube)
  • Put 5 colonies of J06702 in 5mL of LB (one test tube)

Craig and Clay

  • The following parts for limonene synthesis were electrotransformed into the NEB 10 beta strain of E coli: BBa_I742111, BBa_K118024, and BBa_K118025. All DNA was obtained from the distribution plates. These electrotransformed parts were then plated on ampicillin-selective plates and left to grow overnight.
