Observation Transformation
The transformation were observerd on 2pm. Colonies were found for adh2(Z), ADH2(Y) and pirB on the concentrated plates (1300µl spin down discard 900µl, resuspend (200µl) and plated)
Plates without colonies were reincubated at 37°C to check them the next day for potential colonies.
Transfer to LB
Single colonies were picked and inoculated in 5ml LB at 37°C at 250 RPM.
