1. Miniprep of (12)-B, (13)-A, (14)-A, (15)-A, (16)-A, (19)-A solution cultures inoculated from streak plates on 8/30.
Failed due to mistake in procedure; the above numbered parts were again inoculated from streak plates into culture solutions for miniprep tomorrow.
2. Ligation check of (12) through (16), (18) and (19) <- all miniprepped on 8/30; checking was done by taking a small sample for electrophoresis and length comparison.
Results for (14), (18), (19) were not satisfactory; the others seemed okay and may be established as completed parts.
3. Repeat of ligation assembly of (20) and (21): today their constituent parts were treated with restriction enzymes, electrophoresed and purified.
Tomorrow they will be treated with ligation enzymes followed by transformation.
