1. Experimental Group
14:00 sub-culture MG3 with transformed plasmids (A+ & C+) in warm room (37℃), for 120min,
until OD~0.4-0.5.
3mL LB Broth+3μL A++3μL C++60μL bacteria
16:00 blot swimming plates (A+ & C+), using 3μL bacteria.
16:15 put swimming plate into warm room (37℃) for overnight
2. Control Group
14:15 pre-culture un-transformed MG3 (from -80℃) in rotator (32℃) for 2.5h.
5mL LB Broth+ bacteria
16:45 take bacteria out from rotator (32℃)
17:05 sub-culture bacteria in warm room (37℃) for 100min
18:55 take bacteria out from warm room (37℃) and blot swimming plates (without antibiotics)
19:25 put swimming plates into warm room (37℃) for overnight
Suppose: Bacteria on both of these two plates will not swim at all.
