Grow liquid culture to prepare for mini prep
Preparation of Media:
The modified plasmid is kan resistant and the parts pulled from the tool kit are amp resistant, so both media were made.
Kan media:
50 mL LB
50 uL KAN
Amp media:
50 mL LB
50 uL amp
add 3 mL of media to a tube
transfer 1 colony into each tube
shake @ 37 C o/n
