This synthetic biology project will engineer a bacterium from the human gut, Escherichia coli, to detect and fix leaking pipes. Engineered E. coli will detect leaks by homing in on a chemical signal released at the leak site. As the bacteria migrate towards the leak, they will synthesise two proteins, one on the outside of the cell, the other inside, which represent the two components of a biological, protein glue. The adhesive is only activated when the two components mix. The E. coli cells will be engineered to burst at the leak site, mixing the two glue components and creating a sticky protein plug to repair the pipe.

Zussamenfassung  für Laien: Dieses Projekt der synthetischen Biologie manipuliert ein Darmbakterium, Escherichia coli, genetisch so um, dass undichte Stellen in Rohren entdeckt und geschlossen werden können. Genetisch veränderte E.coli sollen diese undichten Stellen finden, indem sie ein chemisches Signal erkennen, das dort ausgeschüttet wird. Während die Bakterien auf diese Stellen zuschwimmen, fangen sie an zwei Proteine zu produzieren; eines ist in die äußere Membran von E.coli verankert und das andere bleibt innerhalb des Backteriums. Diese zwei Proteine representieren die Komponenten eines biologischen Klebers. Wenn sie vermischt werden, wird der Kleber funktionsfähig. Die E.coli Zellen sollen so programmiert werden, dass sie platzen, wenn sie die undichte Stelle erreichen. Dieses Platzen mischt dann die beiden Komponenten und produzieren demnach einen Kleber, der diese Stelle repariert.

তার অংশ হিসেবে আমরা আমাদের প্রজেক্ট তৈরী করেছি মানুষের অন্ত্রে বসবাসকারী Eschrichia coli ব্যাকটেরিয়াকে জীনগতভাবে পরিবর্তন করে এবং এটির কিছু বিশেষ ধর্মকে ব্যবহার করে। আমাদের তৈরি জীনগতভাবে পরিবর্তিত এই ব্যাকটেরিয়া ফাঁটল ধরা পাইপ সনাক্ত এবং পরবর্তীতে সেই ফাঁটল মানুষের সাহায্য ছাড়া মেরামত করতে পারবে। এই প্রজেক্ট সফল হলে এটি দৈনন্দিন জীবনে,পেট্রোলিয়াম কিংবা তৈল শিল্পেই কেবল কাজে লাগবেনা, বরং এই ব্য়াকটেরিয়ার ধর্মকে কাজে লাগিয়ে অষুধ এর সহায়ক উপাদান হিসেবেও ব্যবহার করা সম্ভব যেটি সংশ্লিষ্ট অসুখের বিভিন্ন পার্শ্ব প্রতিক্রিয়া বন্ধ করে কেবল মাত্র নির্ধারিত স্থানে কাজ করতে বাধ্য করবে। আমাদের প্রকল্পের আওতায় বিশেষভাবে পরিবর্তিত এই ব্যাকটেরিয়া দুই ধরণের প্রোটিন তৈরি করবে যার একটি সেল ভেতরে এবং আরেকটি বাইরে অবস্থান করবে। ব্যাকটেরিয়া কেবলমাত্র তখনই ফাঁটল মেরামত করতে পারবে যখন এই প্রোটিন দুটি একসাথে মিশ্রিত হবে।সেক্ষেত্রে বাইরে অবস্থানকারী প্রোটিনটি পাইপ এর ফাঁটল ধরা স্থান সনাক্ত করবে এবং ভেতরে থাকা প্রোটিনটি হবে আঁঠালো ধরণের পদার্থ যা পাইপ এর ফাঁটল বন্ধ করতে সক্রিয় ভূমিকা পালন করবে।
