Today I checked the colony: it grew on both carb and kan, indicating that it was not the intended part. Discovered that PSTI (in FastDigest format from fermentas) must be deactivated by phenol:ethanol precipitation (this is why we didn't get any colonies). Experimented with electrocompetent cells: discovered that they arc at suggested voltage (voltage should be reduced to at least 2000V, 25uF to prevent arcing). -ML
