Colonies containing promoter + RBS + RFP gave some other colonies:

  • Rose colonies: the promoter became constitutive or it strongly expresses, but when you extract the plasmids and do migrations after digestion with EcorRI and PstI, no bands is visible.
  • White colonies: the promoter hasn't mutated.
