Results

  • E. coli cells DB3.1 with the plasmid backbone psB3k3 p1010 (death gene) had one visible colony
  • Positive controls have lots of cell growth
  • No visible colonies on the negative control

Tasks

  • Pick the colony
  • Have individual team meetings
  • Make new LB + Kan plates and broth

Wetlab

  • The one visible colony was picked and placed in 5mL of LB + Kan broth to incubate overnight.
  • The negative control that grew on the LB + Kan June 30, 2009 was replated on two Kan plates
    • one plate was made 6/8/09 and the other was made 7/1/09
      • If there is no growth on the most recent plates, then we know the previous Kan plates have degrated
      • If there is growth, someting is there giving the bacteria a resistence
