9AM, Derek:
DH5alpha for the competent cells are not showing any turbidity yet. Perhaps the bucket of ice in the bath slowed things down too much.
No growth on any of last night's plates.
noon:
still not quite up to the right OD on the cells.
put in 1L of LB to the autoclave and 1L of LB with 15g of agar
3:30:
we made 500ml of Cm plates, 250ml of Amp plates, and 250ml of Km plates
made 250 ml Km broth
picked a colony of P1010Kan and put it in an LB-Kan broth for overnight
prep'ed gels for tomorrow
put cells in fridge because they hadn't quite hit OD and I had to go home.
