Well! We have growth on all the plates: K235009, 13, 14, 16, and 17!

Now I just have to decide when to start a broth culture to get 12-16 hrs...

4.30pm

Broth cultures started at 4.30pm to be taken out at 8.30am tomorrow. I used a new protocol this time: 3 mL Lb + antiobiotic in 5 mL test tubes.

So by Monday we can run a gel and we will know if we have bands. If so, we can talk to Erica about the flow cytometer and see if 13 and 14 are green or not., we should then look in to sequencing. We can also move on with the next assemblies.

Also rehydrated and plated K115017, C0012, and E0240: parts needed for a minimal temperature sensing system.
