1. Conducted miniprep followed by nanodrop concentration check on the following: 10-A, 10-B, 10-D, 10-E.
Results of nanodrop check are as follows:
Conc. 260/280 260/230 10-A 35.2 1.98 1.76 10-B 20.7 1.82 1.59 10-D 37.3 1.90 1.84 10-E 23.7 1.94 1.76
2. Conducted transformation for the following ligated parts: (12), (13), (14), (15), (16), (18), (19), (20), (21).
Also BioBrick part 1-6I was transformed as a positive control.
The plated-out cultures were incubated at 17:30pm.
3. Prepared 42 plates of amphicilin-resistance LA culture medium.
