From 2009.igem.org
We transformed E. coli TOP10 with the following BioBricks (first diluted in 15ul diH2O, and 2ul was taken for transformation)
- gvp (BBa_I750016, Plate 2, well 11A)
- RBS (BBa_B0034, Plate 1, well 2M)
- Terminator (BBa_B0014, Plate 2, well 24C)
- Positive control: pBADnLIC [150ng/ul], 150ug was used
- Negative control: MQ
Incubated for 30min @ ice, 5min @ 37dg, 5min @ ice
800ul LB was added, and in glass tubes they were incubated (while shaking) in 37dg waterbath
100ul was plated on LBA-Amp[100ug/ml] and then the rest was spinned down.
Cell pellet was resuspended and plated.
O/n the plates were incubated @ 37dg.
