1. ligation of (20), (21) vector-insert pairs that were restriction enzyme-treated, electrophoresed and cut out/purified yesterday.
2. miniprep of (12), (13), (14), (15), (16), (19) that were incubated in culture solution yesterday following failed miniprep.
3. transformation of (5), (9), (18), (20), (21) ligated plasmid mixes as the previous attempts at transformation of these plasmids yielded unsatisfactory results.
4. ligation check by restriction endonuclease cut and electrophoresis of plasmids miniprepped this morning.

5. streaked Norad Blue competent cells on agar plate and incubated for preparation of additional transformation-use competent cells.
