Results

Maria and Afton

  • Transformation
    • bacteria are visible on plate (approximately 6 small colonies)
    • no florescence is visible to the naked eye
  • Possible reasons for problems:
    • Promoter works fine: when grown up in overnight culture, RFP was expressed
    • Backbone is there b/c bacteria have Cm resistance and one colony glowed in the past
    • There is no way to see if the RET DNA is there besides running it on a gel
    • Possibly enzymes and buffers for digestion and ligation are not working properly because they have been thawed out and refrozen too many times

Kevin & Adam

  • Early in the morning there was only one small colony visible from yesterday's transformations. At noon there were several more on various plates. Plates will be left all day and possibly overnight before determining the success/failure of yesterday's transformation.

  • Grow up part (Promoter + RET) in overnight culture in LB+AMP selective broth
