Deviated in that we did not concentrate the Synecocystis culture and instead just it straight and at 10 times dilution, corresponding to 10 and 100 times dilution in the protocol.

Triparental mating

Today we put our antisense-RNA constructs in to Synecocystis via triparental mating. Protocol here: Media:Triparental+Mating.txt

Deviated in that we did not concentrate the Synecocystis culture and instead just it straight and at 10 times dilution, corresponding to 10 and 100 times dilution in the protocol.

Evaluating constructs in e.coli

The transformations made yesterday gave plenty of colonies and some were picked and verified in a colony PCR.
