iGEM meeting minutes July 6th
Meeting attendance – Emera will chew off people’s head to get them there.
Science updates: Max: gives to Mitch: We have everything needed to build plasmids to test the reboot trigger. And we’ll be trying to find the inserted piece we couldn’t find. Will try the insertion again too. No colonies in negative control, so by being amp resistant, amp must have recombined in, its just not where it’s supposed to. Doug suggests inverse PCR.
Kalon: plasmids – may be making a new plasmid. PCR team meet after meeting.
Justin: Thursday, updated wiki with gel pictures Primers to beads in, and started PCR for it, but the assay isn’t working. Ordered fancy stuff for Mike’s new method Planning on building plasmid on bead this week. Cloning needed to get screening genes into both plasmids. Need GFP and RFP in pBA and the antibiotic resistance in pAB
Julia: gene order ideas – want to try just doing them in their natural order, then do a segment in a rearranged order. Need to choose which genes to post first. Julia will post the spreadsheets and the handouts from today’s meeting on the wiki
Bioinformatics: looking at media conditions so we can model it. May need to design a bunch of new primers

Business updates: Started corporate fundraising
Would rather not contact U of C and U of L about helping each other. Only need to do one of those 4 choices to get gold – we should definitely ‘develop a new technical standard’, and could easily characterize old biobricks (David will check if the promoters had been characterized…if not we could characterize those).
Human practices: List of possible ideas in Julia’s handout Amber likes pamphlet for high school students idea – meeting Wed at 5.
Workshop this weekend: 6 people are going. Steve is driving too…and there is one other driver (Qiwei?) Emera will be gone wed, so info from Jen Hill will go to David.
