- A09-11 had colonies and satellite colonies
Today
- Ligated and transformed A09-12 (pSB3K3 + ArsR + E0240)
- New PCR to add HA to PCS. Not using a separated cycle instead running 29 cycles at 65C annealing temp
- DNA concentrations of gel extracts of fragments not good enough, hence:
- Amplifying more product for RFP, GFP and rRNA fragments
