  • Midi on RcsB pst1
  • Midi on PAH
  • Mini's (x12) on OP 1-6 and OP+His 1-6
  • Restriction Digest on Dam New 1-8 as sequencing failed 1st time round - wrong DNA sent?!
  • Midi culture set off overnight for CRP-RBS+AK3 colony 1 from replica plate 17/9/09.
  • Housekeeping - tips (all sizes), 1.5ml epis, 2ml epis, and pcr tubes sent to autoclave.

Testing

Harvard-GFP 6th Oct

Protocol considerations

  • Cells were grown overnight in 5ml of M9 media
  • Cells were then diluted and grown at 28 and 42 degrees
  • The cells were allowed to acclimatise to their respective temperatures for 3 hours before the temperature shift took place

Conclusions

  • After 3 hours on ice, the OD stays relatively constant
    • Shows that number of cells during OD measurement and during plating stay the same. Therefore, OD correlates with the number of cells that are plated.
