Results
- Overnight cultures grew well.
Tasks
- Inoculate 2.5 mL from each starter culture into 250mL LB media in 500ml flasks. Culture until ~0.6-1.0 A600.
- Freeze some glycerol seed stocks of NEB10beta and DB3.1 (2 each), concentrate rest 100x and do cold glycerol washes to make competent for electrotransformation.
- Freeze stocks of electrocompetent cells (-80C freezer in George's E. coli box).
Wetlab
- IPTG stock was prepared (2.64mL)
- more IPTG needs to be ordered
- 5uL of IPTG was added to each overnight culture w/ pSB3K3 backbone in E.coli (5mL)
- there were six culture samples
- time for bacteria to begin expressing RFP is unclear
