0800 main culture

divided injection 12.5ml of the culture to 10 50ml centrifuging tubes
WASH 3 times
Add 12.5ml new culture
Centrifuge the 1st tube 3000rpm 5min 20℃ and pick the upernatant
Do same operation every 15min to 2nd~10th tubes
after all, mix the supernatant and solid media at 1:1
make 10 concentration varieties of solid media
