doing the calculation for the Qiagen reagents


one litre of each


buffer P1

50 mM Tris-HCL pH 8.0

10 mM EDTA (MW 292.25 = 2.92g/L)

100 ug/ml RNaseA


[need to know the protocol for pH'ing this buffer... holding off on these calcs]


buffer P2

200 mM NaOH (MW 40 => 8g/L for a 200mM solution)

1% SDS [is this weight per volume? ie. 1% = 1g per 100 ml or 10g per litre? SDS has a molecular weight of 288.38, so in 1L this would be equivalent to a 34mM solution]


-- went hunting for Guanidium, BioBasics was a tenth the price of Sigma but then we found that Caroline Cameron had a 10Kg bucket that we are using.


-- adding NaOH into a small amount of water in a flask, stirring in the fume hood to dissolve. Bringing out of the fume hood to add SDS because the fume hood will blow it around (gloves and masks for the SDS.)


completed 1L of buffer P2, labeled in 100ml bottles.
