following parts were put in competent cells by electroporation:

  • ech; RBS + ech, in pSB1A2,
  • fcs; RBS + fcs, in pSB1A2,

the cells were then put in liquid LB medium to recuperate from the procedure. after a few hours they were transfered on LB/amp plates and grown overnight at 37°C.
