Mixed results from our plates:

K235032 Kan has a few colonies and one colony that is probably mold.

33 Kan has colonies

36 Kan has a few

37 Chl has colonies

05 Chl has many colonies

But J04630 Kan has none. I grew from plasmid this time, and last time it was grown on Amp, perhaps it had lost it's Kan resistance along the way?

2.30pm

Double broth cultures started for K235032 Kan, 33 Kan, 36 Kan and 37 Chl. One broth culture for 05 Chl, and one broth culture (with no antibiotic added because we're almost out of Amp) for J04630 taken from the original Amp plate. One culture of 32 Kan with 3uL 1M IPTG added was also made. Final concentration 1mM.

Does anyone remember what the deal with our IPTG was? I know we had some because I remember using it, but I can't find it. Did it go back to the lab T.A.s or something?

I have made a new stock of 1 M IPTG thanks to Dr. Upton, that's in the -20 freezer, but I would still be interested to know what happened to our old stock.
