08:00 main culture
- divided injection 12.5ml of the culture to 10 50ml centrifuging tubes
- WASH 3 times
- Add 12.5ml new culture
- Centrifuge the 1st tube 3000rpm 5min 20℃ and pick the upernatant
- Do same operation every 15min to 2nd~10th tubes
- after all, mix the supernatant and solid media at 1:1
- make 10 concentration varieties of solid media
