Plasmid prep
A plasmid prep was performed for PRPRYT 3 & 4 on pSB1AK3.
Triparental mating
BG-11 plates were made without antibiotics and without CuSO4 to have the pPetE promoter as silent as possible during triparental mating.
Evaluate constructs in e.coli
The constructs were assembled with the lac promoter (R0011) to make it possible to evaluate them in e.coli, transformed into M15 carrying the lac repressor making it possible for us to induce with IPTG.
