Competent cells, making (again!!)

As our competent cells were again crap (!!) we just have to bite the bullet and make new ones. This time we use a vial of Invitrogen Top10 cells as seed culture.

We also had to make new SOB buffer (as made previoulsy) and in the process made the discovery that our old SOB buffer wasn't pH corrected, it was at pH 6,3 instead of the recommended 7,5! Might very well be the couse of our failed competent cells.
